Occurrence dataset from the 2023 fish distribution survey in waters near Weizhou Island. This dataset comprises a total of 28 orders, 47 families, and 113 fish species.

Sampling Methods

Water samples were collected from eight sampling sites in the waters near Weizhou Island through field sampling. Following environmental DNA extraction, PCR amplification and quantitative fluorescence analysis were performed sequentially, followed by sequencing. After sequencing, taxonomic analysis of ASV representative sequences was conducted using the blastn alignment method. Species information for all fish near Weizhou Island was compiled based on the NCBI database and FishBase database.

Study Extent The study area is located in the waters near Weizhou Island, Guangxi. To be more specific, the scope of the survey is: 21.02 N and 21.08 N Latitude; 109.08 E and 109.15 E Longitude.
Quality Control Purified PCR products were quantified using a Qubit® 3.0 Fluorometer (Life Technologies, Invitrogen). Twenty-four amplicons with distinct barcodes were pooled in equimolar ratios. The pooled DNA was used to construct an Illumina paired-end library following the manufacturer’s genomic DNA library preparation protocol. The amplicon library was then paired-end sequenced (2 × 250 bp) on an Illumina MiSeq platform following standard protocols. Raw sequencing reads were deposited in the NCBI Sequence Read Archive (SRA) under accession number PRJNA1287299.

Method step description:

  1. 1. Collect water samples from eight sampling sites in Weizhou Island waters, with three replicates per site, and photograph field habitats. 2. Extract eDNA, then perform PCR amplification, quantitative PCR, and sequencing. 3. Organize the obtained data and input them into Excel. 4. Conduct preliminary species identification and literature review. 5. To obtain species classification information for each ASV sequence, perform taxonomic analysis of ASV representative sequences using the BLASTn method based on the NCBI database. Compile community composition statistics for each sample at all taxonomic levels: domain, kingdom, phylum, class, order, family, genus, and species. 6. Organize the final data and record the analysis results.
